So maybe I'm stating the obvious, but does that mean the Cash position and 4-2-5 are temporarily shelved until they find a suitable replacement?

No. 4-2-5 is here to stay and will be a main alignment for this defense. KF said it last week and Wallace said the same thing today. There also will be 3-4 with Amani Jones as a "edge defender."

The better question is the 4-2-5 really just a true nickel look or is it somehow different?

The quote says they’ve been practicing at the star (nickel) but doesn’t mention anything about anyone playing cash like Hooker did. How big is the difference?

To me it sounds like as football evolves to more passing and more spread in college especially, Iowa is going to play more nickel coverage, which really isn’t that surprising.

Again I could be wrong but the way I interpret that quote is they aren’t asking DJ or OJ to do as much as what Hooker did. As many of of us estimated last year, Hooker playing cash was more of a product of his versatility, Stone’s emergence, and the attrition at LB.
 
The nickel and the 4-2-5 are different alignments with different personnel in the Iowa defense.

DJ Johnson and Ojemudia are working at the Cash position occupied by Hooker last year.

I didn't listen to the presser, but my guess is the base 4-2-5 will be predicated on having a bit of a tweener in one of the DB slots who is decent in run support and not a liability in pass coverage. It seems like a good adjustment by the staff. The old base 4-3 was great in years where we had at least 2 LBs who were excellent at pass coverage, but years where we lacked that, certain coaches would beat us like a freaking drum.

Kind of like the old Bears Defense was called the 46 defense in honor of Doug Plank who wore #46.

Or hey that's another idea. They could call it the "27" position since that's the number that Hooker wore.

Which is great, but I am more interested as the defense evolves, who can ramp the position up a notch from where Hooker left it. He set the bar pretty high.
